In den vergangenen Jahren hat eine Vielzahl an Studien verdeutlicht, dass die Homöostase und im Speziellen die Adaptation der Skelettmuskulatur an Trainingsreize aber auch an den Einfluss anabol wirksamer Hormonen und Wachstumsfaktoren durch ein komplexes Zusammenspiel molekularer Wirkungsmechanismen moduliert wird. Einige dieser Faktoren, wie z.B. Myostatin, Notch-1, Delta-1 und Sox-8 stehen seit einiger Zeit, bedingt durch Diskussionen um Doping, aber auch altersbedingter Muskeldystrophieerkrankungen, im Blickpunkt des wissenschaftlichen, aber auch des öffentlichen Interesses. Die genauen Zusammenhänge von Muskeldifferenzierung und molekularen Wirkungsmechanismen konnten allerdings noch nicht aufgeklärt werden. Die vorliegende Dissertation untersucht: 1. Den Einfluss von Androgenen und anabolen Steroiden auf molekulare Mechanismen der Satellitenzelldifferenzierung in vitro und dessen Wirkungsmechanismus. 2. Den Einfluss von Training und/oder einer Applikation von Testosteron Propionat auf molekulare Mechanismen der Satellitenzelldifferenzierung in vivo. 3. Die substanz- und gewebsspezifische Wirkung sog. "Designersteroide'' und verschiedener Prohormone in vitro und in vivo. Diesen Fragestellungen wurde mittels molekularbiologischer Analysemethoden wie PCR, Rezeptorbindungstest, Creatin-Kinasebestimmung und Durchflusszytometrie in verschiedenen Trainingsmethoden an Ratten, sowie einem Zellkulturmodell mit murinen C2C12 Myoblasten nachgegangen. Verf.-Referat In recent years, a number of studies have made it clear that homeostasis and in particular, the adaptation of the skeletal musculature to training stimuli and also to the influence of anabolic-acting hormones and growth factors are modulated by a complex interaction of molecular mechanisms of action. For some time, certain of these factors, such as myostatin, Notch-1, Delta-1 and Sox-8, have aroused scientific and also public interest, both because of discussions of doping and because of age-related diseases involving muscular dystrophy. The precise interrelationships of muscle differentiation and molecular mechanisms of action could, however, not yet be elucidated. The present dissertation examines: 1. The influence of androgenic and anabolic steroids on the molecular mechanisms of satellite cell differentiation in vitro and their mechanisms of action. 2. The influence of training and/or application of testosterone propionate on molecular mechanisms of satellite cell differentiation in vivo. 3. The substance and tissue-specific effect of so-called "designer steroids'' and various prohormones in vitro and in vivo. These queries were addressed using molecular-biological analysis methods like PCR, receptor-binding tests, creatin-kinase determination and flow-through cytometry in various training methods in rats, and in a cell-culture model with murine C2C12 myoblasts.
