Skeletal muscle sarcolemmal vesicles from control rats (C) and from rats exercised until exhaustion (E) were used to investigate the effect of a single bout of exercise on lactate transport capacity. Sarcolemmal preparations were not different between the two groups. Exhaustion was associated with a 65% decrease of red gastrocnemius and quadriceps muscle glycogen contents. The time course of 1mM lactate uptake showed that the equilibrium level in group E was significantly lower than group C (p<0.05). The initial rate of lactate uptake at 1 mM increased, with loss of saturation properties. We conclude that a single bout of exercise to exhaustion decreased skeletal muscle sarcolemmal lactate transport capacity in rats, coupled with an alteration in the saturable kinetics at high concentrations. These changes in lactate transport brought about by exercise to exhaustion could alter the metabolism and/or the acid-base equilibrium in the muscle fiber. Verf.-Referat
