Aim: Glutathione-S-transferase M1 (GSTM1) is involved in antioxidant protection and play important role in the removal of reactive oxygen species. Therefore, the aim was to investigate GSTM1 gene polymorphism in young well-trained wrestlers and their relationship with oxidative stress markers following incremental aerobic exercise to exhaustion. Methods: Thirty one young wrestler (age 19.52 +/- 2.75 years, body weight 79.24 +/- 16.13 kg, height 173 +/- 6.49 cm, and body fat 16.37 +/- 5.92 percent) performed an incremental aerobic exercise to exhaustion. Genomic DNA was isolated from whole blood for analyzing GSTM1 gene polymorphism by polymerase chain reaction (PCR). Urine samples were collected before (Pre), after (Post) and 24 h after (24h post) aerobic exercise to exhaustion for analyzing 8-isoprostane and 8-hydroxy-2’-deoxyguanosine, markers of lipid and DNA oxidation. Results and Conclusion: The results demonstrated that the frequency of GSTM1 null and wild-type genotype were 19.35 percent and 80.65 percent, respectively. Wrestlers with GSTM1 null genotype have significantly higher 8-iso PGF2α (Post) and 8-OHdG (24h post) level compared to wild-type GSTM1 genotype (P=0.039 and 0.002, respectively). Furthermore, 8-iso PGF2α and 8-OHdG level significantly increased in GSTM1 null genotype after exercise compared to pre exercise (P=0.039, 0.045); however, there were no significant changes in 8-iso PGF2α and 8-OHdG level in wrestlers with GSTM1 wild-type genotype. In conclusion, GSTM1 null genotype was found to be associated with higher oxidative DNA damage and lipid peroxidation following strenuous exercise in young well-trained wrestlers. Verf.-Referat

Obiettivo: La glutatione-S-transferasi M1 (GSTM1) è coinvolta nella protezione antiossidante e riveste un ruolo importante nella rimozione delle specie reattive dell’ossigeno. Pertanto, obiettivo del presente studio è stato quello di esaminare il polimorfismo del gene GSTM1 in lottatori giovani e ben allenati e la sua relazione con i marcatori di stress ossidativo in seguito a un esercizio aerobico incrementale condotto fino all’esaurimento. Metodi: Trentuno giovani lottatori (età 19,52 +/- 2,75 anni, peso 79,24 +/- 16,13 kg, altezza 173 +/- 6,49 cm e grasso corporeo 16,37 +/- 5,92 per cento) hanno svolto un esercizio aerobico incrementale fino all’esaurimento. Il DNA genomico è stato isolato dal sangue intero per l’analisi del polimorfismo del gene GSTM1 mediante reazione a catena della polimerasi (polymerase chain reaction, PCR). Campioni di urine sono stati raccolti prima (pre), dopo (post) e 24 ore dopo (24 h post) l’esercizio aerobico condotto fino all’esaurimento, per analizzare l’8-isoprostano e l’8-idrossi-2-deossiguanosina, marcatori dell’ossidazione lipidica e del DNA. Risultati e Conclusioni: I risultati hanno mostrato che la frequenza del genotipo GSTM1 nullo e del genotipo wild-type era rispettivamente del 19,35 per cento e dell’80,65 per cento. I lottatori portatori del genotipo GSTM1 nullo avevano un livello significativamente maggiore di 8-iso PGF2α (post) e 8-OHdG (24 h post) rispetto ai portatori del genotipo GSTM1 wild-type (rispettivamente P=0,039 e 0,002). Inoltre, il livello di 8iso PGF2α e 8-OHdG è aumentato in maniera significativa nel genotipo GSTM1 nullo dopo l’esercizio fisico rispetto al livello pre-esercizio (P=0,039, 0,045); tuttavia, non sono state osservate variazioni significative nel livello di 8-iso PGF2α e 8-OHdG nei lottatori con genotipo GSTM1 wild-type. In definitiva, è stato osservato che il genotipo GSTM1 nullo era associato a un maggiore danno ossidativo del DNA e alla perossidazione lipidica in seguito a un esercizio fisico estenuante in giovani lottatori ben allenati.
