Abstract des Autors

Repeatedly biopsy samples can easily be obtained from human skeletal muscles as vastus lateralis, gastrocnemius, tibialis anterior or deltoideus by using percutaneous techniques. By using a conchotome somewhat larger samples can be obtained than by the Bergstroem needle, the sample size can be increased. It is recommended to perform percutaneous biopsies in duplicates to reduce the methodological error. Open biopsies (a surgical technique) may be performed when a large amount of tissue is needed. The biopsy samples may be analysed fresh (e.g. for studies on mitochondria), after freezing (e.g. for fibre typing by histochemistry) or after freezing with subsequent freeze-drying (e.g. for separation of single muscle fibres). To analyse contractile or metabolic characteristics it is possible to choose either a semiquantitative approach using cross-cut frozen tissue for histochemistry (e.g. fibre types) or a quantitative approach by analysing crude muscle homogenates (e.g. enzymes representing oxidative or glycolytic metabolism) or separated cell constituents as mitochondria or myofibrils.
