Abstract
1. Effect of: pH, temperature and ammonia (NH4Cl) have been investigated on the rate of lactate-derived hydrogens utilization via an external pathway in rat skeletal muscle mitochondria. The drop of pH in reaction medium from 7.0 to 6.4 decreased oxidation only slightly (about 10), but the increase of temperature from 22 deg C to 32 deg C enhanced this oxidation about 80. Addition of ammonia to the completely reconstructed system resulted no stimulation of NADH utilization. When NH4Cl was introduced to the medium lacking cytosol, it caused in both pH (pH 7.0 and 6.4) about 45 stimulation of lactate oxidation. 2. The mitochondrial fraction exhibited lactate dehydrogenase activity, which amount was corresponding to 1-2 of the total lactate dehydrogenase activity in muscle homogenate. 3. The mitochondrial LDH was able to feed the external pathway with NADH and this step of NADH oxidation was stimulated by ammonia (NH4Cl). 4. The mitochondrial LDH was involved in direct, coupled to oxidative phosphorylation oxidation of L-lactate by skeletal muscle mitochondria. Verf.-Referat
